ABSTRACT

To clear the effect of the wound to the growth of the larva of the host to the Ophiocordyceps sinensis, the wounds of same severity at the same position were made artificially to the larva and which were artificial fed at the same environment and condition. The results indicated that, over the winter, the survival rate of the wounded of the infection larva was lower than that of the healthy larva, but the weight had no significant difference between the wounded and the healthy larva. The survival rate of the wounded of the no infection larva was lower than that of the healthy larva, but except with black skin, the wounded larva with offwhite and dusty red had no influence on the variety of the weight. In summery, wound had no advantage to the survival rate, but had no influence to the weight. The result had provided theoretical basis to the reforming of the system of the artificial culture O. sinensis.

ABSTRACT

To address the role of Pr1 gene in the process of Hirsutella sinensis infecting Hepialus gonggaensis, differential expression of subtilisin-like protease gene was detected. In the present study, Pr1 gene analogues from H. sinensis were obtained by PCR strategy using specific primers designed from conserved regions of Pr1 gene reported in the GenBank. Then we detected the changes in the expression of Pr1 gene before and after infecting H. gonggaensis using real-time quantitative PCR. We obtained the partial sequence of Pr1 gene with the length of 535 bp (GenBank accession: KC009680). Real-time PCR results showed that the expression level of Pr1 gene was significantly different among 8 samples (P < 0.01). Pr1 gene showed the obvious higher expression level (2-3 folds) after infecting the H. gonggaensis, suggesting that the Pr1 gene may play an important role in the process of H. sinensis infecting H. gonggaensis. The present study paves a way for further identification on infectivity assessment of H. sinensis.

ABSTRACT

<p><b>UNLABELLED</b>To study the effects of the integrated pest control techniques on growth of host larvae of Cordyceps sinensis.</p><p><b>METHOD</b>The integrated pest control techniques were compared with conventional techniques to evaluate the effects on growth of host larvae.</p><p><b>RESULT AND CONCLUSION</b>The results showed that the techniques had broken the balance of the microbial living in the material, produced effective inhibition on the pests, raised the survival rate and promoted the growth of the host larvae at the same time.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the effects of four kind of slow-release fertilizers on yield and quality of Coptis chinensis.</p><p><b>METHOD</b>One to three years C. chinensis was fertilized with slow-release fertilizers twice in April and in September. The yield and nutrient content along with quality of C. chinensis were measured after two years growth.</p><p><b>RESULT AND CONCLUSION</b>All of the slow-release fertilizers increased the yield obviously, and the effect of SRF1 and SRF4 is the most significant. Comparing with control group, the N content in aerial part of 1-2 year-old C. chinensis treated with SRF1 and SRF4 was lower and P and K were higher than that of control group, and the N content in aerial part of 3 year-old C. chinensis treated with SRF1 and SRF4 was higher and P and K were higher than that of control group; The N content in the root of land 3 year-old C. chinensis treated with SRF1 and SRF4 showed no significant difference comparing with control group, and P and K were lower than that of control group, the N and P content in root of 2 year-old C. chinensis treated with SRF1 and SRF4 was higher and K were lower than that of control group. After two years growth berberine content of C. chinensis treated with SRF1, SRF2 and SRF3 were significantly increased, and total alkaloid content of C. chinensis treated with SRF1, SRF3 and SRF4 were significantly increased. We recommend that SRF4 is used as the special fertilizer for 1-year-old C. chinensis, and the SRF1 and SRF4 for 2-year-old C. chinensis, and the SRF1 for 3-year-old C. chinensis.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the dynamic change of the ingredients of Coptis chinensis and evaluate the quality of the crude drugs from main producing areas.</p><p><b>METHOD</b>The ingredients of samples from Shizhu and Wuxi counties in Chongqing, Hongya and Dayi counties in Sichuan, Zhenping county in Shanxi, Lichuan county in Hubei were analyzed for berberine, coptisine, jatrorrhizine, palmatine and the total alkaloids by HPLC and UV methods.</p><p><b>RESULT AND CONCLUSION</b>The contents of measured indexes were mostly highest in 5-years-old C. chinensis. Considering factors such as the yield, it is reasonable to harvest the 5-years-old C. chinensis. There are minor differences in condents of C. chinensis from different areas, all tested samples met the pharmacopoeial standards. It is concluded that the general quality of C. chinensis was good and acceptable.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the changes of nutrient contents in Chinese goldthread soil before and after planting Coptis chinensis.</p><p><b>METHOD</b>The soil in Shizhu, Chongqing, was analyzed for nutrient contents.</p><p><b>RESULT AND CONCLUSION</b>The results indicated that the preplanted soil was strongly acidic and the levels of organic matter, total nitrogen and potassium were high. However, the content of total phosphorus was very low; so were the contents of other eleven nutrients. Variations of nutrients except zinc and molybdenum were large. After four years of planting C. chinensis, the soil pH was increased. Contents of organic matters, manganese, zinc and boron were increased, while the contents of iron and copper were decreased. In general, the variance coefficient of all nutrient elements and the difference of soil fertility were decreased.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the changes of soluble saccharides and amino acids during the growing period of Coptis chinensis.</p><p><b>METHOD</b>Amino acid contents were estimated by Indene tri-ketone colorimetric method; saccharide contents were determined by anthracene ketone method.</p><p><b>RESULT AND CONCLUSION</b>The contents of soluble saccharides and amino acids in different organs were low in plants of 1-2 years old. And they were increased in plants of 3-4 years old. At maturation (5 years), the contents of amino acids kept on increasing in rhizome and flower stalk, while decreased in fibre and leaves, and the contents of soluble saccharides showed the downtrend in rhizome, leaves and fibre, except the uptrend in flower stalk.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To supply the basis of the breeding the host of parasite of Cordyceps sinensis for it's natural culturing, by studying the biology of the host of parasite of C. sinensis, Hepialus biruensis.</p><p><b>METHOD</b>The host of parasite was collected in field, eclosion, mating and spawning in the different conditions were made, and it's biology and the regularity of the growth and development were observed.</p><p><b>RESULT AND CONCLUSION</b>In the natural condition, H. biruensis has the rate of eclosion, 42.3%, and the sex ratio, 1:0.86, and it's lifespan is 2-5 days. It has the rate of eclosion, 67.4%, in the constant temperature. At the 18 degrees C, H. biruensis has the highest rate of mating and spawning, 92% and 98%, and its rate of mating and spawning varies in the different temperature conditions. The male moth cherishes average 814.4 eggs and lays 607.2 eggs. In the open country, the optimum air relative humidity for mating and laying eggs is 75%-90%, while indoors, the optimum air relative humidity is 78%.</p>
